"Summary The present proposal had two main objectives: Objective 1: To perform deep sequencing of sRNAs and degradome libraries from healthy and potato spindle tuber viroid (PSTVd)-infected tomato (cv. Rutger) and N. benthamiana plants to identify, by comparative analyses, host mRNAs targeted by PSTVd-derived small RNAs (PSTVd-sRNAs) possibly involved in pathogenesis Objective 2: To address preliminary steps in the preparation of an EU proposal Two degradome libraries from leaves of tomato plants infected by PSTVd and two libraries from leaves of healthy tomato plants were generated and examined by next generation sequencing (NGS). In parallel, two similar libraries from leaves of N. benthamiana plants (healthy and PSTVd-infected) were also generated and sequenced by NGS. The same RNA preparations used in the degradome sequencing were also used for generating and sequencing, by NGS, the respective cDNA libraries of sRNAs of 21-24 nt. These data have been analyzed and several host genes targeted for degradation by viroidderived sRNAs (vd-sRNAs) have been identified. During the stay of F. Di Serio at IBMCP-UPV, two web-meeting with potential partners in a future EU Call were held. On 22/07/2016 F. Di Serio delivered a seminar at IBMCP in which preliminary data of the activities carried out during his stay at the UPV were presented and discussed with colleagues from the UPV."
