This PhD thesis deals with the development/optimization and validation of a sensitive, accurate, precise and robust LC-MS/MS analytical method for the simultaneous determination of biomarkers of DON, AFB1, FB1, ZEA and OTA in human and animal urine. This method was in-house validated and revalidated in a mini-intercomparison study for human urine. The applicability of the new method to human urine samples was demonstrated in a human pilot study conducted in South Africa and South Italy. The results of this study showed an high percentage of positive samples for biomarkers of DON, FB1, ZEA and OTA in both countries.
Questa tesi di dottorato riguarda lo sviluppo/ottimizzazione e validazione di un metodo analitico LC-MS/MS sensibile, accurato, preciso e robusto per la determinazione simultanea dei biomarcatori di DON, AFB1, FB1, ZEA e OTA. Questo metodo è stato validato con uno studio intra-laboratorio e rivalidato con un mini studio di intercomparazione per urine umane. L'applicabilità del nuovo metodo a campioni di urina umana è stata dimostrata in uno studio pilota condotto in sud Africa e in sud Italia. I risultati di questo studio mostrano un'alta percentuale di campioni positivi per i biomarcatori di DON, FB1, ZEA e OTA in entrambi i Paesi.
