The present study is a summary on the development and inheritance analysis and application of chloroplast microsatellite markers in spatial genetic analysis of the conifer species silver fir (Abies alba Mill.). It aims at discussing and concluding the indicative value of these uniparentally paternally inherited markers in biodiversity and conservation studies at two different spatial scales. When applied among natural populations throughout the natural range the highly polymorphic markers harboured considerable sensitivity towards isolation and/or genetic drift. A pilot study at the stand level revealed the potential of the marker for paternity analysis and reconstruction of the mating system in an endangered relic population. Perspectives are given for a combined usage of DNA markers with contrasting modes of inheritance.
