The availability of suitable protocols for somatic embryogenesis induction in a large number of grapevine genotypes is highly desirable, considering the many applications of this tool. In our laboratory, grapevine somatic embryogenesis has been achieved and studied since 1997, evaluating many Vitis genotypes and particularly V. vinifera cultivars. Up to now, 38 grapevine cultivars and rootstocks have been tested, culturing anthers, ovaries and whole flowers; different media were employed, but most cultures were established on the same medium (PIV originally proposed by Franks et al., 1998). Embryogenic cultures were set up each year, in order to obtain "fresh" material for different purposes. Genotypes with high regenerative competence were identified as well as recalcitrant ones, and subsequently used for further studies on the biological mechanisms underlying the somatic embryogenesis process. The expression dynamics of 12 WUSCHEL (WUS)- related homeobox (WOX) genes present in the V. vinifera genome were analyzed to understand the influence of WOX genes on somatic embryogenesis, comparing their expression profiles in Chardonnay and Cabernet Sauvignon. VvWOX genes appeared to play an important role in the regulation of somatic embryogenesis in grapevine, and the expression of these genes during early phases of somatic embryogenesis differed between the two cultivars.
