we have sought to identify circulating microRNA signatures able to predict MACEs, defined as myocardial infarction (MI), angina, or sudden cardiac death in individuals that had been enrolled in the 60 year olds from Stockholm study. We analyzed 754 microRNAs in the plasma of 100 individuals with major adverse cardiovascular events (MACE) and 100 MACE-free referents, followed for 11 years, to investigate an association with the risk of MACE. We identified 55 microRNAs with significant expression differences between the groups. Among these, microRNA-145-3p was associated with the highest increase in MACE risk (odds ratio [OR]: 2.18), while microRNA-720 was linked to a reduced risk (OR: 0.47). No correlations were found between the microRNAs and levels of C-reactive protein or lipids. Additionally, an interaction analysis revealed 16 pairs of interacting microRNAs, all containing microRNA-320b, whose expression levels influenced the risk associated with the other microRNAs. Higher expression of microRNA-320b correlated with an increased MACE risk associated with the paired microRNAs.
