Photochromic FRET (pcFRET) combines the super-resolution capability of FRET technique with the photomodulation of the optical characteristics of the acceptor chromophore, in view of obtaining a fast and reliable assessment of molecular binding in solution spectroscopy and fluorescence microscopy. This paper summarizes the main photophysical features of pcFRET and describes a model to analyze obtainable data. © 2023 Società Italiana di Fisica. All rights reserved.
