Recent advancements in polymerization strategies and coupling chemistry have enabled the synthesis of complex polymeric architectures with precisely controlled molecular weights and proper functionalities. Star polymers based on amphiphilic polylactide-poly(ethylene glycol) (star-PLA-PEG), with their exclusive structure consisting of multiple variable-length linear chains (arms) radiating from a central core, are biomaterials of growing interest since they combine a three-dimensional spherical and compact shape with a high density of functionalizable end-groups and a covalently reinforced core-shell structure similar to that of micelles.1 In the framework of our studies dealing with the synthesis and functionalization of biopolymers for nanomedicine,2-5 we have synthesized a novel three-arms star-PLAPEG copolymer decorated with β-cyclodextrins (CD) and a novel adamantane-functionalized BODIPY (Ada-BODIPY) to be included by supramolecular interactions into the star-PLA-PEG to monitoring the in vitro cellular uptake and intracellular trafficking of the nanocarrier. The newly designed star-PLA-PEGCD@ Ada-BODIPY was prepared by a proper combination of copper-catalyzed click reaction and hostguest interaction.
