The present study was conducted to explore the effect of culturing peanut (Arachis hypogaea) cotyledons in a novel temporary immersion system, RITA®, for shoot regeneration by caulogenesis. For this aim, 5 combinations of immersion period (8 or 16 min) and frequency of immersion (6, 8 or 16 h) were compared using a liquid MS medium containing BA or TDZ. The tested combinations corresponded to a total daily contact with the liquid culture medium ranging from 12 to 48 min. Best results were achieved with the combination of 16 min of immersion every 16 h which produced a marked increment of shoot forming capacity from peanut cotyledons, in comparison with the culture in semi-solid medium.
