During of the PhD thesis, genotypic and phenotypic differences of Staphylococcus aureus were evaluated. A total of 122 S. aureus strains were isolated from different dairy products (milk, curd, cheese and whey), from different Italian Regions and animal species (cow, goat, sheep and water buffalo). After isolation and purification, the strains were identified and tested for catalase, coagulase, thermonuclease and hemolysis reactions. All 122 strains of S. aureus were tested for SE production by immuno-enzymatic (SEA-SEE) and reversed passive latex agglutination (SEA-SED) methods, while multiplex-PCR was applied for SE genes (sea, sec, sed, seg, seh, sei, sej and sel). Moreover restriction fragments length polymorphism analysis (RLFP) and multiple-locus variable-number tandem-repeat (MLVA) were used to investigate S. aureus diversity in isolates from different areas of Italy.
In questa di tesi di dottorato si sono valutati i polimorfismi fenotipici e genotipici di 122 Staphylococcus aureus provenienti da diverse matrici casearie (latte, cagliata, formaggio e siero), da diverse regioni Italiane e da diverse specie animali (vacca, capra, pecora e bufala). I ceppi in esame, sono stati identificati e caratterizzati valutando la presenza delle attività degli enzimi catalasi, coagulasi, termonucleasi e l´attività emolitica. Successivamente su tutti i ceppi è stata valutata la produzione di tossine mediante metodiche immuno-enzimatiche e genetiche (multiplex PCR). Infine le tecniche RLFP e MLVA) hanno permesso di evidenziare ulteriori diversità genetiche di S. aureus provenienti da varie regioni d'Italia.
